USP <51> Test - Antimicrobial Efficacy Test / USP 51 Preservative Effectiveness Method / AET Test

Antimicrobial preservatives are added to non sterile products including pharmaceutical, cosmetics, personal care products and miscellaneous other antimicrobial products to protect them from microbial contamination.

Antimicrobial efficacy test- United States Pharmacopeia (USP) 51 test and The Cosmetic, Toiletry and Fragrance Association CTFA or Personal Care Product Council, Microbiological Guideline M3 are the test methods to evaluate the effectiveness of an antimicrobial preservative.

Antimicrobial preservative challenge test consists of challenging your product with a high concentration of known microorganisms. According to USP 51 test standard, three bacteria and two fungal strains are used.

Test microorganisms used according to usp 51 test are Staphylococcus aureus, E.coli, Ps.aeruginosa, Candida albicans and Aspergillus brasiliensis (Aspergillus niger).Additional test organisms can also be tested.

Initial concentrations of the test microorganisms are determined by standard microbiological techniques. Microorganism inoculated test products are incubated at 22.5 2.5 C .

Surviving microorganism are counted at 7, 14 and 28 days of the challenge using standard microbiology microorganisms counting techniques. Test microorganisms count and log reduction is calculated at each time interval (7, 14 and 28 days).

Antimicrobial preservative effectiveness of the test product is determined using USP 51 antimicrobial preservative effectiveness test acceptance criteria.
